How to Use Chitosan Bio-Flocculant in Water Treatment

Overview

Chitosan (CAS 9012-76-4) is a naturally occurring cationic biopolymer derived from deacetylation of chitin, the structural material found in crustacean shells (shrimp, crab, krill). With a deacetylation degree of ≥ 85%, commercial water treatment chitosan carries a high density of protonated amino groups (–NH₃⁺) in mildly acidic conditions, giving it a strong cationic charge that enables electrostatic bridging and flocculation of negatively charged particles and colloidal matter.

Chitosan occupies a unique market position: it is the only widely used cationic flocculant that is simultaneously biodegradable, GRAS-listed (FDA 21 CFR 170.30), and approved for use in drinking water treatment and food-contact water by regulatory bodies in the USA, EU (E407a related), and many Asian markets. This makes it the preferred flocculant choice where regulations restrict synthetic polyacrylamide (PAM) or where the dewatered sludge must be land-applied or composted.

How it works: Chitosan's protonated amino groups interact with the negatively charged surfaces of colloidal particles (clay, bacteria, organic macromolecules) via charge neutralization. The polymer chains also bridge between particles, building larger, denser flocs that settle faster than those formed by simple coagulants like alum. At higher molecular weight and dosage, bridging flocculation dominates over charge neutralization.

Preparation & Dissolution

Chitosan powder does not dissolve in pure water — it must be dissolved in a dilute acid solution. 1–2% acetic acid (food-grade vinegar dilution) is the standard solvent for food and water treatment applications. Hydrochloric acid (0.1 M) is used for non-food industrial applications.

Stock solution preparation: 0.5–2% w/v in 1% acetic acid solution.

  1. Prepare 1% acetic acid: dilute glacial acetic acid (or food-grade white vinegar ~5% acetic acid) to 1% v/v in the total water volume
  2. While stirring, slowly sprinkle chitosan powder into the acetic acid solution — do not add all at once as lumps form
  3. Continue stirring for 1–2 hours at room temperature until a clear, viscous solution forms (may appear slightly opalescent at higher concentrations — this is normal)
  4. A 1% chitosan solution in 1% acetic acid has a viscosity of 100–500 mPa·s depending on molecular weight — use a stirring motor, not manual mixing
  5. Filter through 100-mesh screen to remove any undissolved particles before injecting through pumps
  6. Working solution is stable for 3–7 days in sealed containers at ambient temperature; refrigeration (4°C) extends stability to 2 weeks
  7. Do not heat the solution above 50°C — thermal degradation reduces molecular weight and flocculation performance

pH note: Once dissolved, the chitosan solution has a pH of ~4–5. When injected into alkaline water (pH > 7.5), chitosan can partially precipitate as the pH rises above ~6.5. This is usually harmless and the partially precipitated chitosan still flocculates, but very alkaline waters may require pH pre-adjustment or a different chitosan salt form.

Jar test is mandatory: Chitosan dose is highly sensitive to water chemistry (pH, hardness, turbidity type, competing ions). Always conduct jar tests before full-scale dosing. The optimal dose in jar tests is the minimum dose giving ≥ 95% turbidity removal and a settleable floc within 15–30 minutes.

Application Procedure

  1. Prepare chitosan stock solution at 0.5–1% concentration in 1% acetic acid at least 2 hours before the treatment cycle (allow full dissolution).
  2. Conduct jar tests on representative water samples: test 3–5 doses (e.g., 2, 5, 10, 20, 30 mg/L) with rapid mixing (200 rpm, 1 min) followed by slow mix (30 rpm, 10 min) and 20 min settling. Record turbidity and floc characteristics.
  3. For drinking water: verify the selected dose complies with the maximum allowable chitosan residual under local regulations before commissioning.
  4. Commission a peristaltic metering pump (resistant to mildly acidic solutions) at the jar-test-optimized dose rate.
  5. For water clarification: inject into the rapid mix zone (flash mixer or pipe elbow) where turbulence ensures initial charge neutralization, then allow slow mixing for floc growth in the flocculation basin.
  6. For sludge conditioning: dilute chitosan solution to 0.1–0.5% immediately before addition to sludge (thin solutions blend more uniformly). Add at the mixing point upstream of the press or centrifuge feed.
  7. Monitor settled turbidity or floc formation continuously during the first day of operation and adjust dose.
  8. Clean injection lines and pump heads weekly with dilute NaOH (0.5%) followed by water rinse — chitosan can gel in stagnant lines.

Common Mistakes

  • Trying to dissolve chitosan in pure water: Chitosan does not dissolve in neutral or alkaline water. Adding powder to tap water produces an insoluble gel that clogs pumps and lines. Always dissolve in 1–2% acetic acid solution first.
  • Using food-grade white vinegar (5%) without diluting: Full-strength vinegar is 5% acetic acid — this can over-acidify the dosing solution and cause excessive pH drop at the injection point. Dilute to 1–2% acetic acid as the solvent.
  • Skipping the jar test and using a standard dose: Chitosan's optimal dose varies enormously between water sources (from < 1 mg/L for polishing softened drinking water to > 30 mg/L for turbid river water). Never scale up from published dose tables without a jar test on the specific water.
  • Dosing in highly alkaline water without pre-treatment: At pH > 7.5–8.0, protonation of chitosan's amino groups decreases, reducing cationic charge and flocculation efficiency. For alkaline waters, pre-acidify the raw water to pH 6.5–7.0, or combine chitosan with an alum pre-dose that naturally lowers pH.
  • Allowing chitosan stock solution to stand for more than a week: Chitosan solution degrades enzymatically and oxidatively over time, losing molecular weight and cationic charge density. The solution turns from viscous/clear to thin/yellow when degraded. Always check solution viscosity and color before use, and discard solutions older than 7 days.

  • Temperature: 5–30°C, away from direct sunlight and moisture
  • Container: Original sealed polyethylene bags in cartons. Chitosan powder absorbs moisture (≤ 10% moisture spec). Once opened, reseal bags tightly or transfer to airtight containers.
  • Safety: Chitosan is non-toxic, biodegradable, and derived from natural sources. No significant health hazards under normal handling. Powder may cause mild respiratory irritation — use dust mask in dusty conditions. Derived from shellfish (crustaceans) — declare allergen status to operators with shellfish allergies.
