How to Use Glutaraldehyde 50% as a Non-Oxidizing Biocide

Overview

Glutaraldehyde 50% (CAS 111-30-8) is a broad-spectrum non-oxidizing biocide that kills bacteria, fungi, algae, and sulfate-reducing bacteria (SRB) through chemical crosslinking of cellular proteins and nucleic acids. Unlike oxidizing biocides (chlorine, DBNPA), glutaraldehyde is not consumed by organic matter, H₂S, or reducing agents — making it especially valuable in oilfield injection water, gas pipeline systems, and sulfide-rich environments where oxidizing biocides lose their effectiveness rapidly.

The 50% concentration liquid (colorless to pale yellow, density 1.06–1.08 g/mL, pH 3.0–5.0) is effective within 30 minutes of contact. Its biodegradable residual means discharged water containing glutaraldehyde degrades naturally without persistent environmental accumulation. Glutaraldehyde is used in cooling water treatment in rotation with oxidizing biocides to prevent resistance buildup and to penetrate established biofilms that resist chlorine.


HEALTH AND SAFETY WARNING: Glutaraldehyde is a respiratory sensitizer and potential occupational asthma hazard. Repeated inhalation of vapors or mists, even at low concentrations, can cause permanent sensitization — meaning subsequent exposures trigger severe asthmatic reactions. Once sensitized, a worker can never safely work with glutaraldehyde again. Full respiratory protection (minimum OV/P100 half-face respirator), chemical splash goggles, nitrile or neoprene gloves, and chemical-resistant coverall are MANDATORY for all handling operations. Do not perform glutaraldehyde operations in confined or poorly ventilated spaces.


Preparation & Dilution

Glutaraldehyde 50% can be dosed as supplied or diluted for more accurate metering at low doses:

  1. Dilution to working concentration: For cooling water applications (100–300 ppm), dilute 50% glutaraldehyde to 5–10% working solution (1:5 to 1:10 dilution) in clean water. This allows a standard 0–100 mL/min metering pump to achieve ±5% dose accuracy.
  2. Do not dilute below 1% for storage in day tanks — very dilute solutions are susceptible to microbial contamination by glutaraldehyde-tolerant bacteria (especially Burkholderia species) that can degrade the product.
  3. Use only HDPE or stainless steel 316L containers for dilution and day tanks. Copper and brass are not compatible — glutaraldehyde reacts with copper alloys.
  4. Prepare under ventilation: Always dilute glutaraldehyde under a local exhaust ventilation (LEV) hood or with forced ventilation. The vapor pressure at 50°C is significant — never heat glutaraldehyde.
  5. Shelf life of working solutions: Diluted glutaraldehyde (< 10%) has a shelf life of 2–4 weeks at room temperature. Prepare only the quantity needed for that period. Check with litmus — if pH rises above 7, the product is degrading.
  6. Oilfield applications: Glutaraldehyde is often used at full 50% concentration in small-volume metering pumps (glycol-compatible) in oilfield systems. Pre-dilution in oilfield is often not practical — use calibrated chemical injection pumps rated for 50% concentration.

Doses expressed as active glutaraldehyde. For 50% product, multiply ppm by 2 to get mL/m³ of product.

Application Procedure

Cooling water (rotation program):

  1. Establish a biocide rotation schedule: Week 1–2: oxidizing biocide (chlorine/DBNPA); Week 3–4: glutaraldehyde. This rotation prevents development of glutaraldehyde-tolerant biofilm populations.
  2. Dose into the cooling tower basin: Add diluted glutaraldehyde (5–10% working solution) to the tower basin with circulation pump running. Avoid dosing into the hot return water — glutaraldehyde is slightly less stable at temperatures above 50°C.
  3. Allow minimum 4-hour contact time: Do not increase blowdown rate during glutaraldehyde treatment — maintain the residual in the system for the full contact period.
  4. Measure residual by colorimetric test: Use glutaraldehyde test strips or a colorimetric kit (sodium sulfite method). If residual drops below 50 ppm within 2 hours, the system has significant biofilm that is consuming the biocide — shock dose at 500 ppm and consult a water treatment specialist about biofilm removal.

Oilfield injection water:

  1. Inject into the suction of the injection pump: This ensures thorough mixing and distributes the biocide through the injection water before it enters the wellbore.
  2. For SRB control: Monitor H₂S in produced water and injection water weekly. If H₂S > 0.5 mg/L, conduct glutaraldehyde shock treatment at 200–500 ppm. After shock, continue at 50–100 ppm continuous dose.
  3. Temperature limitations: At reservoir temperatures above 80°C, glutaraldehyde degrades faster than the injection fluid travels to the bacteria. At temperatures above 80°C, consider higher doses or alternative biocides (e.g., THPS, tetrakis(hydroxymethyl)phosphonium sulfate).

Common Mistakes

  • Inadequate respiratory protection: The most dangerous mistake. Many workers treat glutaraldehyde like a "safe" liquid biocide because it does not have the immediate irritation sensation of chlorine. But glutaraldehyde's sensitization mechanism is cumulative and silent — a worker may have 50 exposures with no apparent reaction, then suddenly develop permanent occupational asthma on exposure 51. Always enforce OV respirator use and rotate workers to minimize individual exposure time.

  • Using glutaraldehyde alone without rotation: Glutaraldehyde-tolerant bacteria (including some Pseudomonas and Desulfovibrio strains) emerge with exclusive use of any single biocide. A cooling system that uses only glutaraldehyde for months can develop a population shift toward tolerant species, reducing effectiveness. Always alternate with DBNPA, chlorine, or isothiazolinones on a schedule.

  • Dosing at too-low concentration for SRB biofilm: SRBs embedded in established biofilm are protected by the extracellular polymeric substance (EPS) matrix. A continuous low dose of 50 ppm that works well for planktonic bacteria may have zero effect on sessile SRB colonies in biofilm. Shock doses of 300–500 ppm are required to penetrate biofilm and kill embedded populations.

  • Storing diluted product for more than 30 days: Glutaraldehyde undergoes aldol condensation and polymerization over time, especially in dilute solutions and at elevated temperature (> 30°C). Diluted glutaraldehyde that has been stored for more than 4 weeks should be tested for potency before use. A simple colorimetric kit will confirm if the product is still active.

  • Heating or mixing with oxidizing agents: Glutaraldehyde is incompatible with oxidizing agents (chlorine, hydrogen peroxide, permanganate). Mixing causes rapid degradation. Never add glutaraldehyde to a tank that still contains residual chlorine. Always flush systems with fresh water between glutaraldehyde and oxidizing biocide applications.

Storage & Handling

ALL HANDLING OF GLUTARALDEHYDE REQUIRES: OV/P100 half-face respirator (or full face), chemical splash goggles, nitrile/neoprene gloves (minimum), chemical-resistant apron or coverall, and safety boots. No exceptions.

  • Store in original closed containers in a cool (< 30°C), dry, well-ventilated area away from heat and sunlight.
  • Storage vessels: HDPE, SS316L, or FRP (fiberglass-reinforced plastic). Not suitable: copper, brass, zinc, or galvanized steel.
  • Shelf life: 18–24 months at < 30°C in sealed drums away from light.
  • Emergency procedures: In case of skin contact — flush immediately with large amounts of water for at least 15 minutes. Eye contact — flush with water for 20 minutes and seek immediate medical attention. Inhalation — remove to fresh air, seek medical advice.
  • Emergency eyewash and shower required within 10 seconds of handling area.
  • Spill containment: Contain and absorb with dry sand or vermiculite. Do not use organic absorbents. Neutralize with 5% sodium bisulfite solution before disposal. Report significant spills to environmental authority.
  • Transport: Not classified as dangerous goods for most jurisdictions (non-flammable, non-corrosive), but follow SDS shipping guidance.
